Study on Acetylhydrolase Enzyme Activity in Urine of Diabetics
نویسنده
چکیده
The activities of the enzyme acetylhydrolase was studied in urine samples of diabetics and observed to exacerbate diabetic conditions. Acetylhydrolase catabolizes Platelet Activating Factor (PAF) produced by the kidneys by deactivation. Making use of radioimmunoassay technique to evaluate activities in urine samples from non diabetics and diabetics, activity of the enzyme was detected in normal urine with a mean of ±2D = 0.66 nmol/30min/ml. Higher values were however observed among diabetics, specifically relating raised hydrolase activity to this dysfunctional metabolic state in diabetes mellitus.

Urine of patients with nephrotic syndrome contains the plasma type of PAF-acetylhydrolase associated with lipoproteins.
BACKGROUND Platelet-activating factor (PAF) is a proinflammatory phospholipid mediator involved in the pathogenesis of glomerulonephritis (GN). In plasma, PAF is hydrolyzed and inactivated by PAF-acetylhydrolase (PAF-AH), an enzyme associated with lipoproteins, mainly with the low-density lipoprotein. PAF-AH activity has been found in urine of patients with primary GN, however the source and ty...

Introduction & Objective : Telomerase is a reverse transcriptase enzyme that is required in immortalized cancer cells. Early detection of telomerase activity in tumors particularly in bladder cancer is promising. The aim of this study was to compare specificity and sensitivity between cytology and telomerase activity in diagnosis of bladder cancer. Materials & Methods : This study was a case...
متن کاملCharacterization of serum platelet-activating factor (PAF) acetylhydrolase. Correlation between deficiency of serum PAF acetylhydrolase and respiratory symptoms in asthmatic children.
Platelet-activating factor (PAF) acetylhydrolase has been recognized as an enzyme that inactivates PAF. We developed a convenient and reproducible method for determining human serum PAF acetylhydrolase activity. The assay was based on measurement of [14C]acetate produced from 1-O-alkyl-2-[14C]-acetyl-sn-glycero-3-phosphocholine upon precipitation of the complex of radioactive substrate and albu...

Stimulated inflammatory cells synthesize platelet-activating factor (PAF), but lysates of these cells show little enhancement in PAF synthase activity. We show that human neutrophils contain intracellular plasma PAF acetylhydrolase (PLA2G7), an enzyme normally secreted by monocytes. The esterase inhibitors methyl arachidonoylfluorophosphonate (MAFP), its linoleoyl homolog, and Pefabloc inhibit ...
متن کاملPlatelet-activating factor acetylhydrolase in red cell membranes. Does decreased activity impair erythrocyte deformability in ischemic stroke patients?
BACKGROUND AND PURPOSE Platelet-activating factor acetylhydrolase hydrolyzes platelet-activating factor (1-O-alkyl-2-acetyl-sn-glycero-3-phosphocholine). It also hydrolyzes oxidized derivatives of phosphatidylcholine that have a short-chain acyl residue at the sn-2 position. This enzyme may act mainly in the degradation of oxidized phospholipids and may play a role in maintaining erythrocyte de...
